for qualifying a piece of application software, how justifiable is it to set
up a hardware lab to test out compatibility issue?

It really depends upon the app.  With some apps, all that may be required is
passing the Microsoft Logo Hardware Compatibility test (included in MSDN
Professional and Universal subscriptions).  But if you have an unusual mix
of hardware requirements, you probably should commission a competent test
lab.

I'd really need more information before making a specific recommendation.

My new job requires me to test if a number of applications (in-house
and outsourced development) can co-exist on a same platform. the
applications are written in various languages (i.e. it depends on the
app): Visual Basic, Java, PowerBuilder, ...

Does anybody have experience and/or tips in this field? Are there
tools out there to conduct such tests?
