You can do this using c64s:

1. Press F9
2. Press ALT+D(isk)
3. Select your Disk
4. Select a file
5. Press E(xport) and give your new p00-file a name.
6. Press ALT+T(ape)
7. Press N(ew) and give it a name and description
8. Select your new tape
9. Press I(mport)
10.Select you preaviosly exported file.

This is possible, but makes only sense when the program in the D64 is one
file, and not more, since when it's more, it tries to access the drive to
load other parts, and one p00 can't load another.

So it's only possible when the D64 is a sort of archive of standalone files,
not when it is one game spread out over multiple files (and or disks)

Doesn't Trans64 handle that? What about the Star Commander suite?

TRANS64 can copy 6502 binary program files to a T64 'tape'. The files
must first be converted to raw binary before copying.
64 Copy can extract and save in binary format, making it easy to
Import raw files into C64S disk and tape images.

this group (such as I don't)...
